Chimie verte primée dans la fabrication pharmaceutique
Aperçu
Ibuprofen is one of the most consumed non-steroidal anti-inflammatory drugs (NSAIDs) worldwide. The BHC Company (now BASF) developed a revolutionary three-step catalytic synthesis in the 1990s, replacing the original six-step Boots process. This green chemistry approach reduced waste by 99%, achieved near-perfect atom economy, and won the Presidential Green Chemistry Challenge Award in 1997. The process demonstrates how catalytic methods can transform pharmaceutical manufacturing.
Procédé chimique
Isobutylbenzene undergoes Friedel-Crafts acylation with acetic anhydride using HF catalyst, followed by hydrogenation over Pd/C, then carbonylation with CO using Pd catalyst to yield ibuprofen in three atom-efficient steps.
C₁₂H₁₆O + H₂ →[Pd/C] C₁₂H₁₈O
C₁₂H₁₈O + CO →[Pd] C₁₃H₁₈O₂
Matières premières
-
Isobutylbenzene (C₁₀H₁₄) — Petrochemical synthesis (Starting material)
-
Acetic anhydride ((CH₃CO)₂O) — Ketene process (Acylating agent)
-
Carbon monoxide (CO) — Syngas (Carbonylation reagent)
-
Hydrogen gas (H₂) — Steam methane reforming (Reducing agent)
Produits finis
-
Ibuprofen (C₁₃H₁₈O₂) — NSAID analgesic and anti-inflammatory (Racemic mixture, >99% purity)
Environmental Impact
The BHC process achieves 77% atom economy versus 40% for the original Boots process, dramatically reducing waste generation. HF catalyst is recycled with >99.9% recovery. The three-step process generates minimal aqueous waste compared to the six-step alternative.
Considérations de sécurité
- ⚠ Hydrogen fluoride (HF) is extremely corrosive and toxic
- ⚠ Carbon monoxide is toxic and flammable
- ⚠ High-pressure hydrogenation requires specialized equipment
- ⚠ Palladium catalyst handling requires inert atmosphere
Innovations récentes
Asymmetric synthesis methods producing the more active S-(+)-ibuprofen enantiomer are under development, which would halve the required dose.
Continuous flow processes are being implemented to further improve safety and efficiency.
